How to read this certificate

LCMS-UV couples liquid chromatography to a mass spectrometer with an ultraviolet detector. The chromatograph separates the sample into its components over time; the UV detector measures how much light each eluting component absorbs, and the mass spectrometer measures its mass. Purity is reported as the percentage of total UV peak area attributable to the target compound.

Identity confirmation is a separate result from purity. It means the mass measured for the main peak matches the calculated molecular mass of the intended sequence. A sample can be highly pure and still be the wrong molecule — purity and identity answer different questions, which is why both appear on the certificate.

Peptide synthesis varies from batch to batch, so a certificate is only meaningful when it is tied to the specific lot in the vial. The accession number above is the laboratory’s reference for this assay. A generic certificate for a compound, rather than for a lot, describes a different sample than the one you received.

What this certificate does not tell you

UV peak-area percentage is not the same as absolute mass purity: material that does not absorb at the detection wavelength — residual solvent, water, counterion, inorganic salts — is not represented in the figure. These assays also do not test for endotoxin, which is why that line reads N/A. Anyone whose work is sensitive to those parameters should assay for them independently.
